Using in silico bioinformatics, PCR, and sequencing approaches, I am currently interrogating the association of ABC Transporter G family member 2 (ABCG2) with amodiaquine, piperaquine, or lumefantrine resistance. Amodiaquine-resistant, piperaquine-resistant and lumefantrine-resistant Plasmodium berghei (P. berghei) ANKA parasites are being utilized as a surrogate for P. falciparum. Employing Clustal Omega, i am investigating the sequence conservation of the ABCG2 protein across different P. berghei orthologs, and ITASSER to determine ligand binding sites. I am currently mapping non-synonymous mutations by employing PCR and sequencing in the ligand binding regions.
